タイトル Pseudomonas bambusae sp.nov., an Aniline Blue-Decolorizing Bacterium Isolated from Decaying Round Bamboo Culms.

Abstract A novel bacterial strain, designated RC10T, was isolated from decaying round bamboo culms. Phylogenetic and sequence similarity analysis of 16S rRNA gene sequences indicated that strain RC10T was grouped with members of the genus Pseudomonas, with the highest similarity of 98.4% to Pseudomonas lutea DSM 17257T, followed by Pseudomonas kuykendallii DSM 25107T (98.2%). The average nucleotide identity (ANI) and in silico DNA-DNA hybridization (dDDH) values between strain RC10T and species with validly published name of the genus Pseudomonas were less than 84.93% and 20.50%, respectively. Strain RC10T was rod-shaped, Gram-stain-negative, and exhibited optimal growth at 28 °C, pH 6.5, and 0% NaCl (w/v). Notably, strain RC10T showed a high capacity for aniline blue decolorization, with the decolorization rate reaching 93.99% after 48 h of incubation. Genes encoding laccase, DyP-type peroxidases, NADH-dependent FMN reductase, and NAD(P)H dehydrogenase were found in the genome of strain RC10T, which may play crucial roles in aniline blue decolorization. Chemical taxonomic analysis indicated that the polar lipids profile of strain RC10T consisted of diphosphatidylglycerol, phosphatidylglycerol, phosphatidylethanolamine, phosphatidylcholine, three uncharacterized phospholipids, two uncharacterized glycolipids, and three uncharacterized lipids. Summed feature 8 (C18:1 ω7c and/or C18:1 ω6c), summed feature 3 (C16:1 ω7c and/or C16:1 ω6c), and C16:0 was determined as the main fatty acids. Ubiquinone 9 was identified as the sole respiratory quinone. The G+C content of the genomic DNA of strain RC10T was 60.1%. Based on phenotypic, chemotaxonomic, and genotypic features revealed in this investigation, strain RC10T represents a novel species in the genus Pseudomonas, and the proposed name for this novel species is Pseudomonas bambusae sp. nov. The type strain is RC10T (= MCCC 1K08846T = JCM 36624T).
